RESUMO

British × Continental steers (initial BW = 484.6 kg) were fed at a commercial feed yard to evaluate the effects of ß-agonists on live performance, carcass characteristics, and carcass subprimal yield. Weights and ultrasonic measurements were used to allocate steers to pens (n = 40) divided equally into 4 blocks, with 2 treatment replicates per block. Pens were randomly assigned to 1 of 5 treatments: control; ractopamine-HCl (RH) fed at 200 or 300 mg • steer(-1) • d(-1), or 400 mg • steer(-1) • d(-1) top dress for the final 30 d of feeding; or zilpaterol-HCl (ZH) fed at 7.5 mg/kg beginning 23 d before slaughter with a 3-d withdrawal period. Steers were harvested by block at a commercial facility over 4 wk. Carcass based performance measures were calculated using initial pen weights and actual DMI. From each pen, eight carcasses that were within ± 13.6 kg of the mean pen HCW were selected such that two carcasses were within each of the following four Yield Grade (YG) ranges: YG ≤ 2.8; 2.9-3.2; 3.3-3.5; YG > 3.5. Carcasses were fabricated by plant personnel to determine subprimal yield. Steers fed ZH had higher carcass-based ADG and carcass-based G:F compared with all other treatments (P < 0.05). Carcass-based ADG and carcass-based G:F were higher in RH treatments compared with controls (P < 0.05). Steers fed ZH had higher dressing percentages (1.0 to 1.6%) and larger LM area (4.3 to 6.7 cm(2)) than all other treatments (P < 0.05). Use of RH 400 and ZH increased HCW 6.3 and 11.1 kg, respectively compared with controls (P < 0.05). Compared with controls, RH 300 and ZH decreased marbling score and the frequency of carcasses qualifying for upper 2/3 Choice premiums (P < 0.05). Beta-agonists increased subprimal yield from the round and loin; however, blade meat was the only cut from the rib or chuck affected by ß-agonists. Results from this study indicated improvements in performance and carcass traits as a result of ß-agonist use; however, differences between ZH, RH 400, and RH 300 treatments were minimal for carcass traits and cutability. Increases in saleable yield following ß-agonist use were not uniformly distributed across the four major primals and the majority of weight gain occurred in the lower priced cuts of the round and chuck. Increased response of the lower priced cuts to ß-agonists could have economic implications to packers.

The effect of ractopamine hydrochloride (RH) and zilpaterol hydrochloride (ZH) on slice shear force (SSF) and sensory characteristics of beef from calf-fed Holstein steers was evaluated. All steers were implanted with a progesterone (100 mg) plus estradiol benzoate (10 mg) implant followed by a terminal trenbolone acetate (200 mg) plus estradiol (40 mg) implant. Steers were blocked by weight into pens (n = 32) randomly assigned to 1 of 4 treatments: control, RH fed at 300 mg·steer(-1)·d(-1) (RH 300) or RH fed at 400 mg·steer(-1)·d(-1)(RH 400) for the final 31 d of finishing, or ZH fed at 6.8 g/t for 21 d with a 5-d withdrawal before harvest. Fourteen carcasses were randomly selected from each pen, and two LM samples (1 per side) were excised and aged either 14 or 21 d before SSF testing. For trained panel evaluation, two steaks were collected from each of 60 low Choice strip loins (20 each from control, RH 300, and ZH treatments) and aged either 14 or 21 d. Steers fed RH and ZH produced steaks with SSF values that were 9% to 25% higher than controls. No difference in SSF was detected between the two levels of RH (P > 0.05). Compared to controls, the probability of steaks aged 14 d failing to meet SSF requirements to be certified tender (SSF < 20 kg) was increased 0.15, 0.17, and 0.26 in steers fed RH 300, RH 400, and ZH, respectively. Compared to controls, the probability of steaks aged 21 d having SSF values >20 kg was increased 0.03, 0.08, and 0.16 in steers fed RH 300, RH 400, and ZH, respectively. Steaks from Select carcasses of steers fed ZH aged 21 d postmortem had double the probability (0.39 vs. 0.17) of having SSF values >20 kg compared to steaks from steers fed either level of RH (P < 0.05). This difference tended to be identical in steaks from Select carcasses 14 d postmortem (0.50 vs. 0.33; P = 0.11); however, no difference was found in low Choice samples at 14 or 21 d postmortem. Trained panelists rated steaks aged 14 d from steers fed ZH lower for overall tenderness and flavor compared to controls (P < 0.05); however, no difference was found between controls and those fed RH 300. Steaks from steers fed ZH aged 21 d were rated lower for overall tenderness and juiciness compared to controls and those from steers fed RH 300 (P < 0.05). This study suggests RH and ZH negatively impact sensory attributes of beef from calf-fed Holstein steers.

Effects of ractopamine hydrochloride (RH) and zilpaterol hydrochloride (ZH) on saleable yield of carcass sides from calf-fed Holstein steers were evaluated using steers implanted with a progesterone (100 mg) plus estradiol benzoate (10 mg) implant followed by a terminal trenbolone acetate (200 mg) plus estradiol (40 mg) implant. Steers were blocked by weight into pens (n = 32) randomly assigned to one of four treatments: control, RH fed at 300 mg•steer(-1)/d(-1) (RH 300) or RH fed at 400 mg•steer(-1)/d(-1) (RH 400) the final 31 d of finishing, and ZH fed at 60 to 90 mg•steer(-1)/d(-1) (7.56 g/ton on a 100% DM basis) for 21 d with a 5 d withdrawal before harvest. Eight to nine carcass sides were randomly selected from each pen; carcass sides with excessive hide pulls, fat pulls or bruises were avoided. Cutout data were collected within a commercial facility using plant personnel to fabricate sides at a rate of one every 3 to 4 min into items typically merchandised by the facility. All lean, fat and bone were weighed and summed back to total chilled side weight with a sensitivity of ± 2% to be included in the data set. Compared to controls, ß-agonists increased saleable yield of whole-muscle cuts by 0.61%, 0.86% and 1.95% for RH 300, RH 400 and ZH, respectively (P < 0.05). Percent fat was less in carcasses from the ZH treatment compared to controls (P < 0.05); however, this difference was not observed between RH treatments and controls (P > 0.05). Percent bone was less in the ZH treatment due to increased muscle (P < 0.05). The percent of chilled side weight comprised of trimmings was unchanged between treatments, but on a 100% lean basis, RH 400 and ZH increased trim yields (P < 0.05). Analysis of saleable yield by primal showed a fundamental shift in growth and development. Beta-agonists caused a shift in proportion of saleable yield within individual primals, with a greater portion produced from the hindquarter relative to the forequarter, specifically in those muscles of the round (P < 0.05). Beta-agonists increased saleable yield, but these effects were not constant between all major primals. The cutout value gained by packers as a result of ß-agonist use may be influenced more by reduced fatness and increased absolute weight if musculature is primarily increased in the lower priced cuts of the carcass.

Effect of ractopamine hydrochloride (RH) and zilpaterol hydrochloride (ZH) on LM shear force and sensory attributes was determined using pens (n = 40) British × Continental crossbred steers randomly allocated to one of the following treatments: control; RH fed at 200 (RH 200) or 300 mg • steer(-1) • d(-1) (RH 300), or 400 mg • steer(-1) • d(-1) (RH 400) top-dressed for the final 30 d of feeding; or ZH fed at 7.5 mg/kg, beginning 23 d before slaughter with a 3-d withdrawal. Two replicates (pens) per treatment were represented in four blocks. Eighteen carcasses per pen were randomly selected and one 5-cm LM sample was removed from both carcass sides to be used for shear force and sensory evaluation. Samples were aged for 14 d, frozen at -28.8 °C, and cut into 2.5-cm steaks. All steaks were cooked to an internal temperature of 71.1 °C before being evaluated for Warner-Bratzler shear force (WBSF), slice shear force (SSF), or being fed to trained sensory panelists. Increasing dose and potency of ß-agonist increased WBSF by 4 to 17% and SSF by 5 to 24% (P < 0.05). Steaks from steers fed ZH had higher WBSF and SSF values compared with all other treatments (P < 0.05), whereas steaks from controls and steers fed RH 200 were not different (P > 0.05). Probability of steaks failing to meet shear force standards to be certified tender (WBSF <4.4 kg, SSF < 20 kg) was increased from an initial probability of <0.06 in steaks from steers in the control treatment to 0.10 to 0.20 in steers fed RH 400 or ZH (P < 0.05). No difference was detected in panel ratings for overall tenderness of steaks from steers fed RH 200 compared with controls (P > 0.05). Steaks from steers fed RH 300 and RH 400 were comparable for all sensory attributes; however, both RH 300 and RH 400 were rated lower for overall tenderness than controls (P < 0.05). Panelists failed to detect differences in overall tenderness of steaks from steers fed RH 400 and ZH (P < 0.05). Panelists detected no difference in flavor profile or juiciness among treatments (P > 0.05). Results from this study indicated ß-agonists negatively affected beef tenderness and these effects may be more noticeable in steers supplemented with ZH and higher doses of RH.

Fifty-four stores in 30 U.S. cities were sampled from June 2011 through May 2012 to benchmark beef tenderness at retail, as assessed by Warner-Bratzler shear force (WBSF). Top loin (Longissimus dorsi; n = 980) and sirloin (Gluteus medius and Biceps femoris; n = 860) steaks were collected at random (Quality Grade and brand) and shipped via overnight delivery to Colorado State University. From June 2011 through November 2011, North American Beef Tenderness Survey (NABTS) Period 1, samples were shipped fresh and then frozen. Mean WBSF values during Period 1 were 2.9 and 3.5 kg for top loin and sirloin steaks, respectively. Frequencies of steaks classified as tough (WBSF ≥ 4.4 kg) were 8.6% and 17.7% for top loin and sirloin steaks, respectively. When shipped fresh, a disproportionately high frequency (16.9%) of top loin steaks had WBSF ≤ 2.0 kg, representing a deviation from previous works. Two trials were conducted to assess the effect of freezing, retail display, and shipping on WBSF and slice shear force (SSF) of beef top loin steaks. Freezing, retail display, and shipment reduced WBSF by 0.4, 0.3, and 0.0 kg during Trial 1, and by 0.4, 0.3, and 0.1 kg during Trial 2. Slice shear force was lower (P < 0.05) in steaks exposed to shipping conditions during Trial 1; however, this difference was not observed in Trial 2. Shipping decreased the frequency of steaks categorized as tough (SSF ≥ 20.0 kg) from 11.1 to 5.7% and from 30.5 to 28.6%, during Trial 1 and 2, respectively. During Trial 1, WBSF indicated that shipping increased incidence of tough samples from 0.0 to 3.8%, but this trend was reversed during Trial 2 when shipping reduced incidence of tough samples from 13.0 to 5.6%. Coefficients of variation for treatment effects suggested variance remained unchanged (± 2.0%), with respect to shear force values. However, mean values were reduced as a result of shipping conditions. These findings dictated a change in NABTS protocol from December 2011 through May 2012 (Period 2), during which time samples were shipped frozen. Mean WBSF values were 3.4 and 4.0 kg for top loin and sirloin samples, respectively. Frequencies of steaks classified as tough were 14% and 23.5% for top loin and sirloin steaks, respectively. These findings suggest that freezing samples before shipment may influence shear force of steaks collected at the retail level. These data should be considered when evaluating beef tenderness surveys and in the design of future works.

Study objectives were to evaluate ractopamine hydrochloride's (RAC) effect on performance, carcass characteristics, and tenderness of early weaned beef steers. Steers were assigned to a control diet (0 mg RAC·steer⁻¹·d⁻¹), 200 mg RAC mg·steer⁻¹·d⁻¹, or 300 mg RAC·steer⁻¹·d⁻¹. Steers fed 200 and 300 mg RAC·steer⁻¹·d⁻¹ gained 14.84 kg and 14.57 kg more live weight and produced 13.22 and 14.90 kg more hot carcass weight, respectively, than controls. Feed conversions for steers fed 200 or 300 mg RAC·steer⁻¹·d⁻¹ of RAC increased 45.2% and 47.3% and average daily gain increased 55.5% and 54.5% compared to controls, respectively. Feeding either dose of RAC increased (P<0.05) loin muscle area and increased (P<0.05) Warner-Bratzler shear force (WBSF) values compared to controls, however the magnitude of WBSF difference diminished (P>0.05) over 14 days of postmortem aging. Results of this study confirm that RAC increases weight gain and feed efficiency, minimally impacts carcass quality and has manageable impacts on tenderness when fed at either 200 or 300 mg steer⁻¹·d⁻¹.

A beef carcass instrument grading system that improves accuracy and consistency of marbling score (MS) evaluation would have the potential to advance value-based marketing efforts and reduce disparity in quality grading among USDA graders, shifts, and plants. The objectives of this study were to use output data from the Video Image Analysis-Computer Vision System (VIA-CVS, Research Management Systems Inc., Fort Collins, CO) to develop an appropriate method by which performance of video image analysis MS output could be evaluated for accuracy, precision, and repeatability for purposes of seeking official USDA approval for using an instrument in commerce to augment assessment of quality grade, and to use the developed standards to gain approval for VIA-CVS to assist USDA personnel in assigning official beef carcass MS. An initial MS output algorithm was developed (phase I) for the VIA-CVS before 2 separate preliminary instrument evaluation trials (phases II and III) were conducted. During phases II and III, a 3-member panel of USDA expert graders independently assigned MS to 1,068 and 1,242 stationary carcasses, respectively. Mean expert MS was calculated for each carcass. Additionally, a separate 3-member USDA expert panel developed a consensus MS for each carcass in phase III. In phase II, VIA-CVS stationary triple-placement and triple-trigger instrument repeatability values (n = 262 and 260, respectively), measured as the percentage of total variance explained by carcasses, were 99.9 and 99.8%, respectively. In phases II and III, 95% of carcasses were assigned expert MS for which differences between individual expert MS, and for which the consensus MS in phase III only, was < or = 96 MS units. Two differing approaches to simple regression analysis, as well as a separate method-comparability analysis that accommodates error in both dependent and independent variables, were used to assess accuracy and precision of instrument MS predictions vs. mean expert MS. Method-comparability analysis was more appropriate in assessing the bias and precision of instrument MS predictions. Ether-extractable fat percentages (n = 257; phase II) differed among MS (P < 0.05) but were not suitable to predict or validate assigned MS. The performance and reproducibility of expert MS assignment in future evaluations was considered, and an official USDA performance standard was established, to which an instrument must conform to be approved for official on-line MS assessment. The VIA-CVS subsequently was approved to assign MS to carcasses on-line after completion of a 2006 USDA instrument approval trial conducted according to methods developed during completion of this study.

Little information is available regarding the fate of Listeria monocytogenes during freezing, thawing and home storage of frankfurters even though recent surveys show that consumers regularly store unopened packages in home freezers. This study examined the effects of antimicrobials, refrigerated storage, freezing, thawing method, and post-thawing storage (7 degrees C) on L. monocytogenes on frankfurters. Inoculated (2.1 log CFU/cm(2)) frankfurters formulated without (control) or with antimicrobials (1.5% potassium lactate plus 0.1% sodium diacetate) were vacuum-packaged, stored at 4 degrees C for 6 or 30 d and then frozen (-15 degrees C) for 10, 30, or 50 d. Packages were thawed under refrigeration (7 degrees C, 24 h), on a countertop (23 +/- 2 degrees C, 8 h), or in a microwave oven (2450 MHz, 1100 watts, 220 s followed by 120 s holding), and then stored aerobically (7 degrees C) for 14 d. Bacterial populations were enumerated on PALCAM agar and tryptic soy agar plus 0.6% yeast extract. Antimicrobials completely inhibited (p < 0.05) growth of L. monocytogenes at 4 degrees C for 30 d under vacuum-packaged conditions, and during post-thawing aerobic storage at 7 degrees C for 14 d. Different intervals between inoculation and freezing (6 or 30 d) resulted in different pathogen levels on control frankfurters (2.1 or 3.9 log CFU/cm(2), respectively), while freezing reduced counts by <1.0 log CFU/cm(2). Thawing treatments had little effect on L. monocytogenes populations (<0.5 log CFU/cm(2)), and post-thawing fate of L. monocytogenes was not influenced by freezing or by thawing method. Pathogen counts on control samples increased by 1.5 log CFU/cm(2) at d-7 of aerobic storage, and reached 5.6 log CFU/cm(2) at d-14. As indicated by these results, consumers should freeze frankfurters immediately after purchase, and discard frankfurters formulated without antimicrobials within 3 d of thawing and/or opening.

Carcasses that do not conform to mainstream specifications (i.e., those with nonconforming ribeye area) may not achieve their full potential value. Research was conducted to evaluate the relationship between beef carcass LM area at the 12th and 13th rib interface (LMA) and portion size acceptability of other muscles in the carcass. Sixty beef carcass sides of varying LMA sizes (between 67.74 and 116.13 cm(2)) were fabricated to generate 14 individual muscle cuts (triceps brachii long head, infraspinatus, chuckeye complexus, pectoralis profundus, longissimus thoracis, latissimus dorsi, gluteus medius, longissimus lumborum, tensor fasciae latae, psoas major, semimembranosus, biceps femoris, semitendinosus, and vastus lateralis). Retail portion size (g/1.27-cm-thick steak) as well as face surface area and dimensions were recorded for each steak cut perpendicular at the midpoint of the longitudinal axis of each muscle. Subsequently, a nationwide survey was conducted with foodservice chefs and retail meat merchandisers to evaluate acceptability of portion sizes and dimensions of individual muscle cuts. Simple linear regression and nonparametric regression analyses were used to evaluate results of the carcass muscle evaluation and survey, respectively. Results demonstrated that LMA did not affect (P < 0.05) retail portion size of 7 of the 14 muscles (chuckeye complexus, pectoralis profundus, psoas major, semimembranosus, tensor fasciae latae, triceps brachii, and vastus lateralis). Similarly, LMA did not affect (P < 0.05) surface area of steak cross-sectional face areas from 7 of the 14 muscles (chuckeye complexus, psoas major, semimembranosus, tensor fasciae latae, infraspinatus, vastus lateralis, and latissimus dorsi). Muscles for which carcass LMA (P < 0.05) was related to portion size or surface area of portion steaks, or both, were included in the survey. Results of the survey demonstrated that portion size for many muscles were still acceptable to retail merchandisers and foodservice chefs, even though carcass LMA was outside the range of commercially acceptable sizes. Results of this study demonstrated that carcass LMA is not an accurate determinant of the size, and subsequent acceptability, of many other muscles of beef in the carcasses, and may not be a good determinant of value of the beef carcass.

This study evaluated the effects of meat binding or restructuring formulations, including salt/phosphate, algin/calcium, ActivaRM, and Fibrimex, with or without 0.27% (wt/wt) lactic acid, on thermal inactivation of internalized Escherichia coli O157:H7 in ground beef, serving as a model system for restructured products. Ground beef batches (700 g; approximately 5% fat) were mechanically mixed with a 5-strain composite of E. coli O157:H7 (7 log CFU/g) and then with the restructuring formulations. Product portions (30 g) were extruded into plastic test tubes (2.5 x 10 cm) and stored at 4 degrees C (18 h), before heating to 60 or 65 degrees C in a circulating water bath to simulate rare or medium-rare doneness of beef, respectively. Cooking to 60 or 65 degrees C reduced (P < 0.05) bacterial counts of control samples by 1.8 and 3.2 log CFU/g, respectively. Thermal destruction at 60 degrees C was not different (P > 0.05) among all treatments and the control. At 65 degrees C, greater (P < 0.05) thermal inactivation of E. coli O157:H7, as compared to the control, was obtained in samples treated with lactic acid alone (reductions of 4.9 log CFU/g), whereas for all other treatments, microbial destruction (reductions of 2.2 to 4.5 log CFU/g) was comparable (P > 0.05) to that of the control. Cooking weight losses were lower (P < 0.05) in salt/phosphate samples (<1%) compared to other formulations and the control (7.4% to 15.9%). Findings indicated that, under the conditions examined, restructuring of beef with salt/phosphate, algin/calcium, ActivaRM, or Fibrimex did not affect inactivation of internalized E. coli O157:H7 in undercooked (60 or 65 degrees C) samples, whereas inclusion of lactic acid (0.27%) in nonintact beef products enhanced pathogen destruction at 65 degrees C.

A study was conducted to determine if reflectance measurements made in the near-infrared region of the spectrum were additive to reflectance measurements made in the visible region of the spectrum for predicting Warner-Bratzler shear force (WBSF) values. Eighty seven strip loins were collected following fabrication over 3d at a commercial beef processing facility from heifer carcasses with Slight or Traces marbling scores. Spectroscopic measurements were made at approximately 50h postmortem using a Hunter-Lab UltraScan. Subsequently, all strip loins were aged for 14d, cooked to an internal temperature of 70°C, and sheared to obtain WBSF values. Reflectance measurements obtained in the near-infrared region of the spectrum were correlated with WBSF values, however, these measurements were not additive to the predictive ability of reflectance measurements (R(2) values did not differ) made in the visible portion of the spectrum when the use of broad-band wavelength filters were simulated. It was therefore determined, that both the visible and near-infrared spectra measure reflectance and that both methods are acceptable methods of tenderness prediction.

Prion diseases such as bovine spongiform encephalopathy, chronic wasting disease, and scrapie pose serious risks to human and animal health due to a host of disease-specific factors, including the resistance of infectious prions (PrP(Sc)) to natural degradation and to most commercial inactivation procedures. In an attempt to address this concern, a mouse model was used to compare the efficacy of an alkaline hydrolysis process with a simulated continuous-flow rendering treatment for disposal of PrP(Sc)-infected biological material. Female C57/BL6 mice (N = 120) were randomly divided into 4 treatment groups (n = 30), and each mouse was injected intraperitoneally with their designated treatment inoculum. Treatment groups 1 and 2 served as the positive and negative controls, respectively. Group 3 was inoculated with rendered scrapie-positive mouse brain material to investigate the effectiveness of simulated continuous-flow rendering practices to reduce or eliminate PrP(Sc). Group 4 was inoculated with hydrolyzed scrapie-positive mouse brain material to determine the sterilizing effect of alkaline hydrolysis on PrP(Sc). Mice were monitored for overt signs of disease, and those showing clinical signs were killed to prevent undue suffering. Brains were obtained from all mice that died (or were killed) and analyzed with an ELISA for the presence of PrP(Sc). Results indicated that the simulated continuous-flow rendering treatment used for preparing the rendering treatment group inoculum failed to completely eliminate PrP(Sc). Rendering delayed, but did not stop, clinical mouse-adapted scrapie transmission. Compared with positive controls, the rendering treatment group experienced an approximate 45-d average delay in days to death (250 vs. 205 d for positive controls; P < 0.0001) and a death loss of 73.9% (P = 0.0094). Positive controls suffered 100% death loss. The results validated the efficacy of the alkaline hydrolysis treatment to inactivate all PrP(Sc) because no alkaline hydrolysis treatment group mice succumbed to the disease (P < 0.0001). Based on our results, alkaline hydrolysis should be considered by the animal rendering and beef packing industries as an alternative to incineration, landfill burial, and rendering for disposing of biological material potentially infected or contaminated with prion disease.

Transportation of cattle to the slaughter plant could influence hide contamination with Salmonella enterica. Fecal and hide samples were obtained from 40 lots of cattle at the feedlot and again at the slaughter plant. Potential risk factors for hide contamination were evaluated. A multilevel Poisson regression model was used to determine whether transportation and lairage were associated with hide contamination by Salmonella. Cattle with hide samples positive for Salmonella at the feedlot had twice the risk of having positive slaughter hide samples compared with cattle without positive feedlot hide samples (relative risk [RR], 1.9). Cattle transported in trailers from which samples positive for Salmonella were collected had twice the risk of having positive slaughter hide samples compared with cattle transported in culture-negative trailers (RR, 2.3). Cattle transported for long distances had twice the risk of having positive hide samples at slaughter compared with cattle transported shorter distances (RR, 2.3). Cattle held in lairage pens contaminated with feces had twice the risk of having positive slaughter hide samples compared with cattle held in clean pens (RR, 1.8). Cattle held off feed longer than 18 h before loading had twice the risk of having positive slaughter hide samples compared with cattle held off feed for shorter times (RR, 1.7). Cattle that were agitated during loading had twice the risk of having positive slaughter hide samples compared with cattle that were calm (RR, 2.2). These findings suggest that variables associated with transportation and lairage can impact the presence of Salmonella on the hides of cattle at slaughter.

This study evaluated the fate of inoculated Listeria monocytogenes on frankfurters stored under conditions simulating those that may be encountered between manufacturing and consumption. Frankfurters with or without 1.5% potassium lactate and 0.1% sodium diacetate (PL/SD) were inoculated (1.8 +/- 0.1 log CFU/cm(2)) with a 10-strain composite of L. monocytogenes, vacuum-packaged, and stored under conditions simulating predistribution storage (24 h, 4 degrees C), temperature abuse during transportation (7 h, 7 degrees C followed by 7 h, 12 degrees C), and storage before purchase (60 d, 4 degrees C; SBP). At 0, 20, 40, and 60 d of SBP, samples were exposed to conditions simulating delivery from stores to homes or food establishments (3 h, 23 degrees C), and then opened or held vacuum-packaged at 4 or 7 degrees C for 14 d (SHF). Pathogen counts remained relatively constant on frankfurters with PL/SD regardless of product age and storage conditions; however, they increased on product without antimicrobials. In vacuum-packaged samples, during SHF at 4 degrees C, the pathogen grew faster (P < 0.05) on older product (20 d of SBP) compared to product that was fresh (0 d of SBP); a similar trend was observed in opened packages. At 7 degrees C, the fastest growth (0.35 +/- 0.02 log CFU/cm(2)/d) was observed on fresh product in opened packages; in vacuum-packages, growth rates on fresh and aged products were similar. By day 40 of SBP the pathogen reached high numbers and increased slowly or remained unchanged during SHF. This information may be valuable in L. monocytogenes risk assessments and in development of guidelines for storage of frankfurters between package opening and product consumption.

The National Beef Quality Audit-2005 assessed the current status of quality and consistency of US fed steers and heifers. Hide colors or breed type were black (56.3%), red (18.6%), Holstein (7.9%), gray (6.0%), yellow (4.9%), brown (3.0%), white (2.3%), and brindle (1.0%). Identification method and frequency were lot visual tags (63.2%), individual visual tags (38.7%), metal-clip tags (11.8%), electronic tags (3.5%), bar-coded tags (0.3%), by other means (2.5%), and without identification (9.7%). Brand frequencies were no (61.3%), 1 (35.1%), and 2 or more (3.6%), and brands were located on the butt (26.5%), side (7.4%), and shoulder (1.2%). There were 22.3% of cattle without horns, and the majority of those with horns (52.2%) were between 2.54 and 12.7 cm in length. Percentages of animals with mud or manure on specific body locations were none (25.8%), legs (61.4%), belly (55.9%), side (22.6%), and top-line (10.0%). Permanent incisor number and occurrence were zero (82.2%), 1 (5.2%), 2 (9.9%), 3 (0.4%), 4 (1.2%), 5 (0.1%), 6 (0.3%), 7 (0.0%), and 8 (0.7%). Most carcasses (64.8%) were not bruised, 25.8% had one bruise, and 9.4% had multiple bruises. Bruise location and incidence were round (10.6%), loin (32.6%), rib (19.5%), chuck (27.0%), and brisket, flank, and plate (10.3%). Condemnation item and incidence were liver (24.7%), lungs (11.5%), tripe (11.6%), heads (6.0%), tongues (9.7%), and carcasses (0.0%). Carcass evaluation revealed these traits and frequencies: steer (63.7%), heifer (36.2%), bullock (0.05%), and cow (0.04%) sex classes; dark-cutters (1.9%); A (97.1%), B (1.7%), and C or older (1.2%) overall maturities; and native (90.9%), dairy-type (8.3%), and Bos indicus (0.8%) estimated breed types. Mean USDA yield grade (YG) traits were USDA YG (2.9), HCW (359.9 kg), adjusted fat thickness (1.3 cm), LM area (86.4 cm(2)), and KPH (2.3%). The USDA YG were YG 1 (16.5%), YG 2 (36.3%), YG 3 (33.1%), YG 4 (11.8%), and YG 5 (2.3%). Mean USDA quality grade traits were USDA quality grade (Select(90)), marbling score (Small(32)), overall maturity (A(64)), lean maturity (A(57)), and skeletal maturity (A(68)). Marbling score distribution was Slightly Abundant or greater (2.7%), Moderate (4.3%), Modest (14.4%), Small (34.5%), Slight (41.2%), and Traces or less (2.9%). This information helps the beef industry measure progress and provides a benchmark for future educational and research activities.

Transportation of cattle from the feedlot to the slaughter plant could influence hide contamination of Escherichia coli O157. A study was initiated to investigate the influence of transportation and lairage on shedding and hide contamination of E. coli O157. Fecal and hide samples were obtained from 40 pens of harvest-ready beef cattle at the feedlot prior to transport and again at the slaughter plant immediately after slaughter. Potential risk factors for hide contamination at the feedlot, during transport, and at slaughter were evaluated. A multilevel Poisson regression model was used to evaluate if transportation and lairage were associated with hide contamination by E. coli O157 in finished beef cattle. Lots of cattle held in E. coli O157-positive lairage pens had eight times greater risk of having positive slaughter hide samples compared with cattle held in culture-negative pens (relative risk, 8.0; 95% confidence interval, 1.6 to 38.8). Lots of cattle that were held in lairage pens contaminated with feces had three times greater risk for positive slaughter hide samples compared with cattle held in clean pens (relative risk, 3.1; 95% confidence interval, 1.2 to 7.9). Lots of cattle that were transported for long distances (> 160.9 km) had twice the risk of having positive hide samples at slaughter compared with cattle transported a shorter distance (relative risk, 2.4; 95% confidence interval, 1.1 to 5.1). These findings suggest that transportation and lairage should be considered in E. coli O157 control strategies.

The removal of 18,345 specified risk materials was observed during audits of 18 U.S. beef processing facilities that, in total, account for over 90% of total U.S. beef slaughtered. Audited plants varied in capacity (280 to 6,000 head per day) and processed both "fed (young cattle)" and "nonfed (mature cows/bulls)" cattle. When all observations for removal of specified risk materials were combined from plants and adjusted for type of cattle processed, overall compliance with specified risk material removal regulations was 98.08%. A 100% compliance rate for removal of brains and distal ileums was recorded based on a total of 600 observations for removal of brains and a total of 2,400 observations for removal of distal ileums. Observations for removal of dorsal root ganglia were collected from 16 of the 18 plants, and overall compliance for dorsal root ganglia removal was 99.6% (4,783 of 4,800). Fifteen of the 16 plants were 100% compliant. For tonsils, data from 18 plants were collected, and tonsils were correctly removed from 92.8% (4,777 of 5,145) of tongues and heads. Data for spinal cord removal were collected from 18 plants, and the spinal cord was removed completely in line with U.S. Department of Agriculture-Food Safety and Inspection Service regulations for 99.43% of the observations. Based on the results of this study, packing plants have demonstrated that they are complying with regulations for removal of specified risk materials from beef meat products intended for human consumption greater than 98% of the time. To continue to assure food safety and consumer confidence, continued vigilance and provision of training programs for plant workers are essential.

Effects of the dark-cutting condition were examined on commercially slaughtered beef carcass sides that were classified into groups exhibiting 1/3, 1/2, and full degrees of the dark-cutting (DEGDC) condition, as evaluated by a USDA-Agricultural Marketing Service grader (n = 20 per group). Twenty-nine muscles of each carcass side were evaluated to determine the ultimate pH and color (L*, a*, and b*). Fourteen beef muscles (biceps femoris, deep pectoral, chuck complexus, gluteus medius, infraspinatus, latissimus dorsi, psoas major, longissimus thoracis, longissimus lumborum, semimembranosus, semitendinosus, triceps brachii long head, tensor fasciae latae, and vastus lateralis) were evaluated using Warner-Bratzler Shear force (WBSF) and a trained sensory panel. The muscle x DEGDC interaction was significant for ultimate pH, L*, a*, and b* values (P < 0.05). When ultimate pH values of individual muscles were compared with the same muscles evaluated in a previous study, the 1/3, 1/2, and full DEGDC had 7, 9, and 5 muscles, respectively, that fell within a computed 95% prediction limit of what would be considered as a normal pH but were more variable as measured by within-class CV. Color values (L*, a*, and b*) of the muscles from dark-cutting carcasses were numerically lower than those from the normal carcasses. A survey designed to determine the ideal color range of beef lean for retail meat merchandisers (n = 34) and food service chefs (n = 33) across the United States resulted in data analyzed using principal components analysis of L*, a*, and b* values for muscles dissected in the study to estimate the true values for dark-cutting carcasses. Muscles that were within an acceptable color value range for food service chefs had the potential to add between $42.29 to $26.44 and $14.71 to $8.11 per side when valued at Choice and Select prices, respectively. Muscles that were within an acceptable color value range had the potential to add between $30.39 to $16.74 and $10.37 to $5.03 per side for retail meat merchandisers when acceptable muscles were valued at Choice and Select prices, respectively. No muscle x DEGDC interactions were detected for WBSF and sensory panel scores (P > 0.05), but differences were detected among muscles (P < 0.05). Several muscles were considered salvageable from the dark-cutting carcasses that were evaluated, and no significant differences in sensory scores or WBSF between DEGDC classes suggested equal sensory expectations for muscles from dark-cutting carcasses.

Four experiments were conducted in commercial beef-packing facilities The objectives of these experiments were to: (i) determine and validate a carcass sampling technique and location to determine if central nervous system (CNS) cross-contamination exists/occurs; (ii) determine if residual CNS tissue contamination remains on splitting saws after sanitation procedures; (iii) determine the prevalence of CNS cross-contamination in commercial slaughter facilities; (iv) determine whether washing treatments reduce or eliminate CNS tissue presence in carcass-splitting saws; (v) determine the effectiveness of commercial spray-washing systems in removing CNS tissue from beef carcasses; and (vi) compare residual CNS tissue levels on the blade and in the housings of the Jarvis Buster IX and Buster IV carcass-splitting saws. CNS tissue remained, albeit at very low levels, in the housings and on the blades of carcass-splitting saws after carcass splitting and operational sanitation. Additionally, after splitting carcasses, CNS tissue remaining in the splitting saw housings and on saw blades was found to cross-contaminate subsequent carcasses during splitting. Most splitting saw operational sanitation procedures reduced the amount of CNS tissue remaining in the splitting saw housings and on splitting saw blades, but no treatment eliminated CNS tissue from either to levels below the detection limit of the assay (6 ng/100 cm2). Washing in carcass spray-washing cabinets at three of the five commercial beef-packing facilities reduced, but did not eliminate, presence of CNS tissue in the aitch bone area of carcasses. Carcass spray washing in cabinets at three of the five facilities reduced (P < 0.05) the concentration of CNS tissue in the fourth thoracic vertebra area. While extremely low concentrations of CNS tissue remained in the splitting saw housings, on the splitting saw blades, and on carcasses, it is unknown whether these levels would pose a human food safety risk because the exact amount of bovine spongiform encephalopathy-infected spinal cord capable of transmitting the disease to humans is dependent on the infectivity titer, which is not readily known.

The objective of this study was to model with logistic regression the growth/no growth interface of different initial inoculation levels (10(1), 10(3) and 10(5) CFU/ml; study 1), or nonadapted vs acid-adapted (study 2) Escherichia coli O157:H7 as influenced by pH, NaCl concentration and incubation temperature. Study 1 was conducted with a mixture of four E. coli O157:H7 strains grown (35 degrees C, 24 h) in tryptic soy broth (TSB). Study 2 was conducted with the same mixture of four E. coli O157:H7 strains grown (35 degrees C, 24 h) in glucose-free TSB with 1% added glucose (final pH 4.83), or in diluted lactic acid meat decontamination runoff fluids (washings; final pH 4.92), or nonadapted cultures prepared in glucose-free TSB (final pH 6.45), or in water washings (final pH 6.87). Parameters included incubation temperature (10-35 degrees C), pH (3.52-7.32), and NaCl concentration (0-10% w/v). Growth responses were evaluated for 60 days turbidimetrically (610 nm) every 5 days in 160 (study 1) and 360 (study 2) combinations in quadruplicate samples, with a microplate reader. The lower the initial inoculum the higher were the minimum pH and a(w) values permitting growth. Differences in the pH and a(w) growth limits among inoculum concentrations increased at 15 and 10 degrees C. Acid-adapted cultures were able to grow at lower pH than nonadapted cultures, while at temperatures below 25 degrees C, growth initiation of nonadapted cultures stopped at higher a(w) compared to acid-adapted cultures for the whole pH range of 3.52 to 7.32. A comparison with available data indicated that our model for acid-adapted E. coli O157:H7 in different environments may provide representative growth probabilities covering both nonadapted and stress-adapted contaminants.
